Evaluation of DNA Binding Affinity with Polymer Supported Peptides |

It is important to identify chromosomal pattern which affects biological mechanism such as cell growth or cell signaling by modulating protein expression. In this study, peptide sequences which are related in DNA binding were synthesized by Solid Phae Peptide Synthesis (SPPS) method. Here we tried to synthesize peptides on solid supports for efficient screening the sequence specificity of binding DNA. The beads were incubated in the solution of fluorescently labeled DNA and then the peptide sequences of the beads were analyzed by MALDI-TOF MS. In the future, this method will be used to determine unknown peptide sequences binding specific DNA fragments. |
