Inthis study, guanidine-modified cationic hydroxyethyl cellulose (HECP2kG) wasmade for efficient gene delivery system. Hydroxyethyl cellulose (HEC) is acompetent candidate for drug delivery carrier due to its water solubility andbiocompatibility. Guanidine moiety which is known for its cellular penetratingability can be utilized as an effective tool for enhancing cellular uptake of thegene carrier. Cationic hydroxyethyl cellulose (HECP2k) was synthesized byreductive amination of low molecular weight polyethylenimine (PEI, Mw 2000) tooxidized HEC. HECP2k was further modified by guanidinylation with DIPEA and1HPC, resulting in HECP2kG. Physicochemical properties of HECP2kG/pDNA polyplexwere characterized while cytotoxicity and transfection efficiency were examinedby MTT assay and luciferase transgene expression assay. Hence, gene deliveryefficiency of HECP2kG was studied in comparison with HECP2k in vitro. |
