Premature drug release is a limitation of many polymer micelle drug carriers. Chemical conjugation of drugs to polymers through labile linkages is a promising method for addressing this challenge. In this paper we demonstrate a simple approach for achieving such a carrier system. A hydrophobic cancer drug, doxorubicin, was chemically conjugated to the hydrophobic (i.e., PnBA) portion of a PEG-PnBA block copolymer through hydrazone bonds. A hydrazone link is stable under physiological (neutral) pH conditions, but it degrades at low pH (such as in acidic regions of tumors, or in some intracellular compartments). Micellization and degradation properties of this doxorubicin-grafted block copolymer have been characterized under varying pH conditions. The pH-dependent chemotherapeutic properties of this carrier system have been investigated in vitro using HeLa cells. |
