hMSC using for CaP/dopa-HA gene delivery by inducing osteogenic differentiation |

Human mesenchymal stem cells (hMSC) are one of the stem cells that have multipotent properties. However, use of hMSC is still limited to clinical steps. Genetic modification of hMSC is known to provide an opportunity of controlling cell type as well as their behavior. Herein, osteogenetic major protein of cDNA with cloning methods was presented to the human body to induce bone differentiation. An organic-inorganic hybrid gene delivery system supports more efficient differentiation. Calcium phosphate (CaP) gene delivery system was designed. The dopa-modified hyaluronic acid (HA) and growing CaP nanoparticles were adsorbed to act as both a gene vector and a particle stabilizer. CaP/dopa-HA nanoparticles showed significantly higher transfection in hMSCs because of the specific HA-binding to CD44+ expressed on their cell surface. CaP/dopa-HA/BMP2-pDNA achieved the desired gene expression. This study represents successful osteogenic differentiation of hMSC. |
