제목 Fabrication of bio-glass modified injectable bone substitute based on polyelectrolyte complex for improved bio-compatibility and controlled drug delivery
초록 In this work we fabricated a modified injectable bone substitute (IBS) for improved biocompatibility with the addition of bio-glass powder with conventional TTCP/DCPA based bone cement. The unique feature of physical cross linking of cationic and anionic polymers to form a polyelectrolyte complex (PEC) was exploited to enhance the biocompatibility of IBS cement. We used chitosan as cationic polymer. Alginate and gelatin was used as anionic polymers. Alginate was modified by oxidizing to improve the solubility. On the other hand a cross linking agent was used with gelatin for a chemical cross linking between chitosan and gelatin. The PEC was used as a polymer modifier for the bone cement and 20% solution of the cationic and anionic polymers were employed. Parameter like setting time was evaluated with addition of different content of bio-glass powders. The IBS was incubated for different time at 37 °C temperatures and was evaluated for the formation of HAp. The fabricated IBS was characterized for its morphology by SEM. Mechanical properties like Vickers hardness was evaluated. To investigate the bio-compatibility we performed the MTT assay and investigated the cell proliferation and adhesion behavior by cell culturing on the IBS for 1, 3 and 5 days and by one cell morphology.
