Preparation of DNA Block Copolymers and Amphiphiles via Solid-Phase and Enzymatic Methods |

This presentation provides an overview of hybrid biopolymers namely lipid DNAs and DNA block copolymers (DBCs) which made of abasic liphophilic nucleotides and nucleic acids covalently conjugated to polymers, respectively. Solid-phase and enzymatic synthetic methods for such materials will be discussed in detail. On one hand, solid-phase synthesis merits coupling of hydrophobic segments to synthetic oligonucleotides through standard detritylation coupling using conventional DNA synthesizer, which yielded lipid DNAs and amphiphilic DBCs. On the other hand, DBCs could be engineered by nucleic acid enzymes such as polymerase and ligase for the preparation of ultra-high molecular weight multiblock DBCs. Owing to the artificial segments (i.e. polymers and lipids), the hybrid materials possess additional self-assembly abilities in addition to the programmability of DNA. A few examples for the application of materials will be outlined. |
