Probably, the most familiar, widespread and useful medical device is suture. Therefore, if a surgical suture is used as a gene delivery platform, it can be a convenient and user-friendly method. However, previously reported gene vector immobilized surgical sutures have several drawbacks, which include low transfection efficiency and uncontrollable release. To overcome these problem, sutures for gene delivery were fabricated using the electrostatic interaction between chitosan and heparin. As chitosan is electrostatically cationic, heparin is chosen for an anionic natural polymer. The reason for choosing heparin is that it binds with various proteins especially adeno-associated virus which has been widely used in a variety of gene therapy trials due to its safety and efficacy. Thus, heparin has been used as an AAV reservoir for storage and controlled release. |
