Therapeutic Effects of TGF-β/LMC Conjugates on the Osteoarthritis in vivo |

It has been reported that TGF-β is related to biological activity, such as cartilage specific matrix formation. Also, it has been demonstrated by previous our study that half-life of growth factors can be prolonged by conjugating with LMC(low molecular weight chitosan, Mw :10535). Based on these reports, we prepared TGF-β-LMC conjugates by using carbodiimide, and investigated its effects on articular cartilage defects of the rabbit's knees. Cartilage defect was made at rabbit's knees, and after 3 days TGF-β-LMC conjugates was injected into one side and PBS was injected into the other side. Experimental group showed significantly positive staining(type II collagen) on the extracellular matrix around the chondrocyte-like cells of articular defect. consequently, it is suggested that TGF-β/LMC conjugates can be used as a therapeutic agent for Osteoarthritis. |
