Controlling cell-matrix and cell-cell interactions is critical for regulation of cellular activities, such as cell adhesion, proliferation, and differentiation. Many studies have attempted to improve cell adhesion ability of synthetic matrix by modification with ECM molecules, growth factors, or other bioactive molecules. However, controlling cell-cell interactions of synthetic matrix for stem cell differentiation is still lacking. In this study, D1 stem cells were cultured on either RGD peptide- or LRP5 peptide-modified alginate hydrogels. RGD peptide-modified alginate was used to investigate the cell-matrix interactions, and LRP5 peptide-modified alginate was used to investigate the cell-cell interactions. LRP5 peptide-modified gels were useful to enhance chondrogenic differentiation of D1 stem cells even at the low cell density, compared to RGD-modified alginate gels. |
