제목 Study on the degradation behaviour in vitro for PLGA as drug carrier

Degradable polymers have been the focus of extensive research because of the potential for long-term unattended therapy and site-specific drug administration without the need for device retrieval. Among the degradable polymers currently under investigation, poly(DL-lactide-co-glycolide) (PLGA) copolymers are the most widely studied because of their long history of safe clinical use as drug carrier. 50:50 PLGA was used as a model degradable polymer in this study to investigate the degradation behaviour on drug release from bulk degradable polymers in vitro. 1,3-bis(2-chloroethyl)-1-nitrosourea(BCNU) as water non-soluble drug and 5-flourouracil (5-FU) as water soluble drug were used as model drugs. Molecular weight change, residual mass, water uptake, morphological change of wafer, and pH of release test media were characterized to investigate the effect of polymer degradation on drug release. Release of BCNU from PLGA wafer and 5-FU from PLGA wafer followed almost zero order release kinetics and almost tri-modal release kinetics respectively.
