제목 Controlled Release of Bovine Serum Albumin from Hydroxyapatite Encapsulated within Hyaluronic Acid Hydrogels
초록 A novel sustained release formulation of bovine serum albumin (BSA) was developed using hydroxyapatite (HAp) and hyaluronic acid (HA) hydrogels. BSA was complexed with hydroxyapatite (HAp) through electrostatic interaction between Ca2+ of the HAp and COO- of BSA. For the preparation of HA hydrogels, adipic acid dihydrazide grafted HA (HA-ADH) was synthesized and analyzed with 1H NMR. The degree of HA-ADH modification was ca. 69%. BSA/HAp complex was in situ encapsulated into HA-ADH hydrogels using bis(sulfosuccinimidyl) suberate (BS3) as a selective crosslinker. The denaturation of BSA during HA-ADH hydrogel synthesis was drastically reduced with decreasing pH from 7.4 to 4.8. The specific reactivity of BS3 to hydrazide at pH = 4.8 might be due to its low pKa (pKa = 3.0) compared with that (pKa > 9) of amine of BSA. In vitro release of BSA in PBS at 37℃ could be controlled from two days over a week and will be discussed in detail.
