Physiological and environmental characteristics of succinate-producing Actinobacillus succinogenes sp. 130Z were studied to develop the efficient biological system for succinic acid production. Fed-batch cultures and continuous cultures were examined to develop the biological system for high-productivity succinic acid production. Several factors which affect cell growth and succinic acid production were examined by flask and batch cultures. Other carbon sources including lactose, sucrose, maltose, fructose, mannitol and mannose could be utilized as alternatives to glucose. Among the eight different complex nitrogen sources, only soytone could be utilized for succinic acid production. Fed-batch cultures were examined using various feeding strategies. By constant feeding, cells grew up to OD660 of 6 and succinic acid productivity was 1.17 g/L/h. however, the productivity decreased as culture time went on. By intermittent feeding, cell concentration couldn't reach OD660 of 4. Continuous cultures at various dilution rates were also examined. |
