Optimization of Corsslinked Polyvinyl Alcohol-Gelatin Hydrogels to Obtain High Mechanical Properties and Excellent Biocompatibility for Bone Regeneration |

The aim of this investigation was to optimize crosslinking conditions of polyvinyl alcohol/gelatin hydrogels using genipin as naturally crosslinker for bone application. To satisfy implantation, compressive strength, materials characterization, biocompatibility and biodegradation were investigated. Ratios of polyvinyl alcohol/gelatin:genipin (1:1) offered excellent compressive strength (wet: 8 MPa, lyophilized: 20 MPa) among series ratios. Crosslinking reaction of gelatin and genipin during synthesis process was reflected by fourier transform spectroscopy. Crosslinked hydrogels were non-toxic with over 90 % cell viability in 100 % extract solution using MTT testing. Biocompatibility testes showed excellent cell attachment and cell proliferation using osteoblast cells (MG-63), chondrocytes cell and mesenchymal stem cells (MSC). Protein expression of beta-actin, osteoglycine and PCNA showed that the crosslinked hydrogels were non-harmful for cell membrane and nucleus using western blot testing. Crosslinked hydrogels started to degrade at day 10th and totally degraded after 3 month in which satisfy to shorten healing process. |
