제목 Synthesis and characterization of an electrospun membrane/ hydrogel  novel bilayer scaffold for skin tissue regeneration
초록 A novel bilayer scaffold composed of electrospun PCL/ PLGA (80:20) membrane and glutaraldehyde (3%) crosslinked chitosan: gelatin hydrogel (1:3) was fabricated using two methods: by overlaying and casting method (BS_1) and by electrospinning of membrane to the lyophilized hydrogel (BS_2). The fabricated scaffolds were examined by SEM for gross morphology and were characterized by its mechanical strength, porosity, biodegradation rates and water uptake ability. Biocompatibility of the materials was investigated by cell adhesion and cell proliferation studies by MTT assay, western blot detection of β-actin protein and SEM micrographs. All tests were compared to a separate hydrogel and membrane scaffold alone. Experimental results showed that the structural characteristics of the bilayer scaffold can support tissue engineering functions and displays good cell-material interaction. Moreover, it was found out that in combining the two biomaterials the properties of one distinct layer can complement the other layer to make an effective tissue engineering scaffold.
