Molecular Design, Expression and Characterization of Thermally Responsive Multiblock Polypeptides for Enhanced Cell Penetration |

Enhanced drug delivery to target cells by using cell penetration domains is important for advanced drug delivery. We are specifically interested in thermally responsive elastin-based polypeptides (EBPs) and cell penetrating peptides (CPPs) as advanced drug delivery carriers. The EBPs are thermally responsive biopolymers with lower critical solution temperature behavior for their self-assembly while the CPPs making translocation through the plasma membrane efficient. In this study, thermally responsive multiblock polypeptides composed of EBPs, CPPs and Cys domains were genetically engineered and purified. Their self-assembled micelle structures and chemically stabilized micelles via disulfide bonds were characterized by UV absorption and dynamic light scattering as a function of temperature. This work suggests that the thermally responsive multiblock polypeptides could open a new avenue for molecular design of advanced drug delivery systems with enhanced cell penetration. |
