Synthetic regulators have been widely utilized to control the expression of gene in microbial cells, and riboswitches are well used input part of synthetic regulators. For diverse applications of the synthetic regulator, tuning of dose-response parameters is important. However, the direct tuning of riboswitches was constrained by their difficulties and narrow range of modulation. Therefore, a new modular part is required that can tune the dose-response parameters with ease. Here, we developed a toehold switch-based genetic modulator. Combination of previously reported hybrid input part, which use riboswitch and transcriptional factor, and toehold switch showed dramatic improvements in the dynamic range and operational range. Furthermore, the modularity of the toehold switch was also demonstrated by using a different inducible promoter system. This strategy is applicable for tuning other biosensor systems and could be widely implemented in metabolic engineering and synthetic biology. |
