제목 Cancer targeting and imaging using genetically assembled double-layered fluorescent protein nanoparticles

Real-time image-guided surgery recently has attracted considerable attention in surgical cancer treatment, which depends on intraoperative imaging techniques to guide the surgical procedure. For intraoperative imaging, fluorescence techniques can visualize specific cancer tissues are important. Hepatitis B virus capsid (HBVC) which is a self-assembled protein nanoparticle comprised of 180 or 240 subunit proteins can be used as fluorescent proteins. The HBVC is a kind of cage for genetic encapsulation of fluorescent proteins (FPs). This self-quenching of FPs can be controlled by the spacing between fluorescent proteins. The genetically modified HBVC with double layers of mCardinal fluorescent proteins and cancer cell receptor binding peptides (mC-DL-HBVC) exhibit a igh fluorescence intensity, and is efficiently internalized by cancer cells and stable against intracellular degradation. Additionally the mC-DL-HBVC can detect tumor in live mice campared to a conventional fluorescent dye, Cy 5.5. This suggests the great potential of mC-DL-HBVC as a promising contrast agent for in vivo tumor imaging.
