Induction of Cardiac-Lineage Primed hMSCs via Nanoporous Polymer Membrane Co-Culture for Myocardial Infarction Treatment |

Human mesenchymal stem cells (hMSCs) have been considered as a potential treatment for heart diseases such as myocardial infarction. In particular, previous studies show that cardiac-lineage primed hMSCs are known to possess better therapeutic efficacy than naïve hMSCs for myocardial infarction. For the first time in this line of research, we developed a novel stem cell co-culture platform based on biocompatible, highly porous, and ultrathin polymer membranes to control the hMSC behavior toward cardiac lineage by the co-culture of hMSCs with cardiomyoblast cell line (H9C2). After 1 week of co-culture, the cardiac lineage commitment of hMSCs was assessed, showing significantly higher levels of connexin43 and cardiac structural/ion channel biomarkers in the MSCs co-cultured with well-defined nanoporous and ultrathin polymer membranes. |
