제목 | Heparin conjugated Polymeric Micelle for Controlled Delivery Carrier of Hydrophobic Drug and Growth Factor |
초록 | Polymeric micelle (PMs) using amphiphilic block copolymers have been widely studied due to their potential application in drug delivery system as they are capable of forming aggregates in aqueous solutions. However, they have some limits for the enhanced therapeutic effect on the target sites, which can be required the multi-actions of drugs to use. Here, we developed new functional PMs by introducing heparin molecules into biodegradable backbonefor dual controlled release. This heparinized PMs can be interacted various growth factors such as bFGF, VEGF, TGF-1and BMP as well as incorporating hydrophobic drug in a core. Tetronic-PCL (TC) amphiphilic block copolymers were synthesized by ring opening polymerization of -caprolactone in the presence of Tetronic using stannous octate as a catalyst. TC was carboxylated with succinic anhydride and then carboxylated TC was coupled with heparin by EDC and NHS chemistry. Indomethacin (IMC) loaded PM was prepared by a solution evaporation method. Briefly, IMC in methanol was added to Tetronic-PCL-Heparin (TCH) solution. Spontaneous formation of micelle containing solution went through evaporation step, allowing evaporation of methanol and kept overnight at 25℃. Free IMC was removed from PMs solution by centrifugal separation. bFGF was added to IMC loaded PM solution and stirred 3 hrs at 21℃. Free bFGF was removed by adding PEO solution (10 wt%) using salting-out salt method. Salts was removed by centrifugal separation and then lyophilized. The synthesized copolymer was characterized by FT-IR, 1H-NMR and GPC. The immobilized amount and the bioactivity of heparin were estimated with toluidine blue assay and APTT test. Drug loaded PM was characterized by TEM, fluorescence spectroscopy, dynamic light scattering and UV-visible spectroscopy. bFGF quantitative assay was carried out using human FGF basic immunoassay. The objective of this studyis to develop antiphlogistic drug and growth factor loaded heparinized PMs, which have thehydrophobic drug in a core and the heparin binding growth factor in an outer shell as a multi-functional drug for pharmaceutics and tissue engineering. |
