제목 Intervertebral Disc Regeneration using SIS Scaffolds
초록 Objective: Intervertebral disc degeneration causes back pain and disability. A solution of the problem developed using biomaterials such as polymer and ceramic scaffold. Tissue engineering technique using biodegradable polymer, cytokine and cell might be supported very effective and viable organ. Small intestinal submucosa (SIS), which was natural materials improvement attachment, proliferation and differentiation. In study, we experimented 3D of disc cell of using SIS scaffold.



Method: We manufactured SIS 1%, EDC 100mM sponge for using scaffold, and cultured disc cell from rabbit was seeded 1x105 cells/scaffold concentration. Then it designed Four groups such as (1) monolayer cultured group in growth media and chondrogenic media included TGF-β1 as positive control (2) cultured group growth media in SIS scaffold, and (3) in chondrogenic media TGF-β1 in SIS scaffold. (4)cultured group growth media in PLGA scaffold as negative control. After seeded cell, we observed charcterization of disc cell by MTT assay, RT-PCR on 3, 7, 14 and 21 day.



Result: In MTT result, we observed that chondrogenic media better than growth media for cell proliferation. RT-PCR results, we confirmed more express type Ⅱ collgen and aggrecan in chondrogenic media.



Conclusion: when we cultured disc cell in vitro, we observed better to keep the characteristic of disc cell in 3D than that in 2D culture, and more stable to culture disc cell in chondrogenic media than that in growth media.
