In this study, a novel poly(lactic-co-glycolic acid)(PLGA) scaffold were prepared by paraffin sphere leaching for bone regeneration. Then two type of scaffold, glycol chitosan immobilized on PLGA(GCH-PLGA) scaffold and BMP-2 peptide immobilized on GCH-PLGA(BMP-2-GCH-PLGA) scaffold were prepared via the crosslinking agents reactive carboxyl groups on its surfaces introduced from alkali treatment for increasing hydrophilicity, cell attachment and proliferation. The incorporating of bone morphogenetic protein peptide (BMP-2) promoted osteoinduction in Gel-Pec-BCP was developed for bone induction. The physical properties were examined by scanning electron microscopy (SEM), porosity, Fourier transform infrared spectroscopy(FT-IR) and X-ray Diffraction (XRD). In vitro experiment was done by studying of cytotoxicity using MTT assay, and immunefluorescence for cell behaviour. In vivo study was done by implantation in nude mouse model, it is concluded that our modified scaffolds hold a great candidate for bone tissue engineering application. |
