제목 Development of novel naringenin riboswitches that enable naringenin dependent translation-initiation regulation
초록 Ligand-dependent genetic regulation enables facile control of the pathway gene expression and screening of competitive producer strains. Transcription factor-based sensors are often accompanied by the off-target effect and delayed response which results from trans-regulatory mechanism. On the contrary, riboswitch could bypass the aforementioned issues. Here we demonstrate the process of developing riboswitches which specifically sense the intracellular concentration of naringenin by modulating the concentration of supplemented flavonoids and antibiotics in the selection process. Selected riboswitches from few rounds of toggled selection work at designed operational ranges. Finally, we validated the specificity of the riboswitches for naringenin against other flavonoids with a common flavonoid backbone. We expect that these new riboswitches could be applied to diverse engineering to screen optimal strain or monitoring metabolites.
