Ion releasing nanoparticles controlling the osteogenic cellular microenvironment in human mesenchymal stem cells |

In this study, we have developed a metal ion-releasing nanoparticle for enhanced osteogenic differentiation of human mesenchymal stem cells (hMSCs). In order to enhance the osteogenic differentiation and induce angiogenesis of hMSCs, zinc and iron were synthesized simultaneously into a nanoparticle with pH-sensitive degradation property. Our studies revealed that treating optimized concentration of our nanoparticles to hMSCs delivered zinc and iron ion in a controlled release manner and supported early osteogenic gene expression with improved angiogenic paracrine factor secretion. Concurrent intracellular release of zinc and iron ions through the endocytosis of the nanoparticles further stimulated the mild reactive oxygen species generation level in hMSCs without cytotoxicity and thus improved the osteogenic capacity of the stem cells. Current results suggest that our ion releasing nanoparticles might support the future biomedical applications for bone regeneration. |
