제목 | Reduction of nonspecific binding by using a new linker and anti-β-galactosidase in the interferometric biosensor |
초록 | An interferometric sensor using immobilized recombinant E.coli has been developed for a rapid and easy detection of endocrine disruptors. When recombinant E.coli GW1040(dinD::lacZ) are exposed to DNA damaging compounds, they releases β-galactosidase by a SOS regulon system. Very low enzyme activities less than 100 unit-ml-1 responding to μg-ml-1(ppb) level of endocrine disruptors can be easily determined by this interferometric biosensor which can solve the problems in a conventional enzyme assay such as several time-consuming procedures and tedious cell collection. Heavily doped p-type porous silicon was fabricated to produce a Fabry-Perot fringe pattern, which caused effectively the change in the refractive index of the medium including β-galactosidase. An interferometric sensor reacts in a dose-dependent manner with the shift of Fabry-Perot fringe pattern in response to endocrine disruptors. Both the stability in surfaces of porous silicon by modifying with prolinker®-A instead of a typical biotinylation and the reduction of nonspecific bindings by using anti-β-galactosidase have been obtained in this study. |
