제목 Biodegradable Temperature Sensitive MPEG-PCL Copolymer Hydrogels
초록 Temperature-sensitive polymer have been investigated extensively for the drug delivery system and tissue engineering. Especially, biodegradable thermo-reversible hydrogels has many characters such as good solubility in water, easy drug loading, non-toxicity, and biodegradability. Recently PEG-based diblock and triblock polyester copolymers, stimulating to temperature, were studied as injectable drug delivery and tissue regeneration.1,2
In this study, to prepare thermo-reversible sol-gel polymers, we synthesized the diblock copolymer consisting of methoxy polyethylene glycol (MPEG) and ε-caprolactone (ε-CL) by ring opening polymerization in the presence of HCl․Et2O at room temperature. The concentrated PBS solution of MPEG-PCL diblock copolymers changed from sol to gel phase as the temperature increased. Also, we observed protein release of 20 wt% PBS solutions loading 0.1 % bovine serum albumin, dextran, and insulin. Hydrogels containing proteins showed the sustained release behavior without the initial burst. To confirm the gel formation at body temperature, we observed the mice after injection of 20 wt% aqueous solution of block copolymer. After two months, we confirmed the gel on mice without the dispersion of gel.
In conclusion, we confirmed that these MPEG-PCL diblock copolymers have possibility for the biomedical applications as injectable drug delivery carrier and tissue engineering via in situ gel formation.
