Synthesis, Characterization and Glutathione-Mediated Drug Release of Poly(ethylene glycol)-b-Poly(s-butyl cysteine) Containing Disulfide Linkage |

A diblock copolymer, poly(ethylene glycol)-b-poly(s-butyl cysteine) containing disulfide bond was synthesized and characterized by 1H NMR, Fourier transform infrared (FT-IR), and gel permeation chromatography (GPC). The critical micelle concentration (CMC) of copolymer was 3.8ⅹ10-6 M in water. Indomethacin (IMC) as a model drug was incorporated into the micelle by an oil/water emulsion method. The diameters of polymeric micelle without and with the entrapped IMC were measured to be ca. 75.8 and 95.7 nm, respectively. The drug release from drug loaded polymeric micelle was investigated as a function of glutathione concentration in the release medium. The release study revealed that the release IMC from polymeric micelle was increased by increasing the gluthathione concentration and indicated that the release IMC from polymeric micelle can be controlled by adjusting glutathione concentration, demonstrating their potential as drug delivery vehicles. |
