제목 Release and characterization of double-walled microspheres containing water soluble drugs
초록 When administering drug into the body, it reveals the decline of drug effect or toxicity. This is due to the concentration of the drug administered in vivo. One method, which is to impregnate drug into polymer matrix, has studied for the control release of drug.1-3 To overcome these problems, double-walled microspheres studied with biodegradable polymers had been investigated. 5-Fluorouracil (5-FU) as water soluble drug has been used widely for one of antimetabolite anticancer however, ratio of oral absorption is very low(25%).4 In this study, We manufactured double-walled microspheres using oil-in-water(O/W) solvent evaporation method with/without ultrasonication to increase the encapsulation ratio of 5-FU and to control the release of 5-FU, and to prevent side-effects.
Double-walled microsphere is composed of two components; Core consists of the mixture of dextran and 5-FU and corona consists of PLGA. Double-walled microsphere was characterized by scanning electron microscope (SEM) observation, confocal fluorescence laser microscope observation, and HPLC to measure the release behavior and the encapsulation ratio of 5-FU.
