The bioavailability of drugs such as sildenafil and celecoxib can be limited by their poor dissolution into aqueous body fluids following administration. With using polymer-directed crystallization technique, crystalline particles of sildenafil and celecoxib having controlled release patterns, improved processability, and stability were developed. Crystalline particles were prepared in presence of various polymers (poly(vinylpyrrolidone), hydroxypropyl methylcellulose, and Pluronic F-127) using ethanol and water as a solvent and a non-solvent, respectively. The characteristics of crystalline particles were characterized by differential scanning calorimetry (DSC), X-ray diffraction (XRD), IR spectroscopy and scanning electron microscopy (SEM). Polymer contents and crystallization temperature have prominent effects on the solubility and morphology of particles. The present study shows that polymer-directed crystallization is effective in controlling the in vitro release of drugs. |
