제목 Induction of chondrogenic differentiation of adipose-derived stem cell(ASC) using F127-based hydrogel
초록 Human adipose-derived adult stem cells (ASC) have the ability to differentiate into a chondrogenic phenotype in three-dimensional culture and media containing dexamethasone (Dex) and transforming growth factor (TGF). The purpose of this study is to make a Pluronic-based hydrogel, in which hydrogel can carry a specific functionality toward the induction of chondrogenic differentiation of ASC without exogenously added chondrogenic supplements. In this work, a Pluronic (F127,F) derivative (FGM) was synthesized using glycolide (G) and 4-methacryloxyethyl trimellitic anhydride (META, M). To add functionality, Dex and heparin (Hep) are grafted to the Pluronic derivative,producing FGM-Hep and FGM-Dex. For in vivo test, ASC was mixed with Pluronic-based hydrogels containing TGF and Dex. The results of nude mouse implantation for 4 weeks suggested that functional Pluronic-based hydrogel has a potential as an injectable system in stimulating chondrogenesis of ASC.
