Chondroitin sulfate (Cs) is an acidic mucopolysaccharide found in bone, cartilage, and connective tissue. It can form an ionic complex with positively charged transforming growth factor-beta1 (TGF-β1). The purpose of this study is to make a nanoparticle (TGF-ß1/Cs) that can promote the chondrogenesis of human adipose-derived stem cells (hASC). The morphology and size of fabricated ionic complex was determined nanoparticle using SEM and DLS. To identify the complexation of TGF-ß1 and Cs, this nanoparticle using was characterized by circular dichroism (CD) and zeta potential. For the efficacy of this nanoparticle for chondrogenesis of hASC, nanoparticles are encapsulated in fibrin gel with hASC. The cell-loaded hydrogels are cultured in 12-well plates for 4 weeks in vitro. The effect of nanoparticle is evaluated using histological staining, GAG contents, and RT-PCR for cartilage-specific gene expression. |
