Cartilage tissue engineering using TGF-β1 loaded functional PLGA nanoparticles and a elastic PLCL scaffold |

In cartilage tissue engineering, the chemical and mechanical stimulations are very important for differentiation and maintenance of the lineage. Also, 3-dimensional spatial organization of cells is crucial to functional cartilage formation. We formed substantial tissue engineered cartilage with human adipose derived stromal cells (hADSCs) and TGF-β1 loaded functional nanoparticle–hydrogel-Polymer complex. We used mechano-active scaffolds of poly(lactide-co-caprolactone) (PLCL) having a complete rubber-like elasticity and functional nanoparticles made with poly(lactide-co-glycolide) (PLGA) as a hydrophobic core, Pluronic F-127 as a hydrophilic surface layer, and heparin as the functional moiety. The hybridization of a gel and a PLCL scaffold and TGF-β1 loaded nanoparticles with continuous controlled release can encourage hADSCs to differentiate to chondrocytes, maintain their phenotypes and enhance GAGs production and consequently, improve the quality of cartilaginous tissue formed. |
