GRGD-conjugated enzymatic in situ formation of hydrogel from gelatin-4-hydroxylphenylacetamide and oxidized alginate loaded biphasic calcium phosphate as injectable scaffolds |

Hydrogels consisted of the Glycine-Arginine-Glycine-Aspartic (GRGD)-conjugated oxidized alginate and gelatin-4-hydroxylphenylacetamide was obtained by enzymatic crosslinking under physiological conditions using horseradish peroxidase (HRP) as a catalyst and hydrogen peroxide (H2O2) as an oxidant. The gel formation was varied within a time period of 34 to 91 seconds by controlling the concentration of polymers. The hydrogel structure was highly porous and showed unique channelized morphology with an extensive branching of the channels. The physical properties and the degradation of hydrogel were confirmed by water uptake and degradation test. In addition, the hydrogels showed a good cytocompatibility with rat bone marrow stem cells confirmed by MTT assay and live-dead assay. These hydrogels could be promising injectable biomaterials as a regenerative cell matrix and the drug delivery systems. |
