Peripheral artery disease (PAD) is a disorder that is induced mainly by atherosclerosis and thrombosis and restricts the blood flow. Therapeutic angiogenesis has been recently considered as a promising therapy for patients with PAD. In the ischemic site, reactive oxygen species (ROS) such as hydrogen peroxide (H2O2) are generated in large amounts. H2O2 is one of essential oxygen metabolites in living organisms, but its overproduction of H2O2 leads to oxidative stress. In this work, we synthesized H2O2-responsive PVAX nanoparticles which encapsulate curcumin as a synergistic agent in ischemic injuries. Curcumin-loaded PVAX nanoparticles (CUR-PVAX) generate CO2 bubbles in the site of ischemic injury which could significantly enhance ultrasound signal and release vanilly alcohol and curcumin which have antioxidant, anti-inflammatory and angiogenic effects. Therefore, CUR-PVAX nanoparticles hold tremendous potential as a theranostic agent for ischemic diseases such as PAD. |
