제목 TGF- β2/PD98059-Loaded Porous Scaffold to Enhance Chondrogenesis  and Suppress Hypertrophy from Stem Cells
초록 Many researchers believed that growth factor-immobilized 3D porous scaffolds may be a good alternative to enhance the cartilage regeneration using mesenchymal stem cells (MSCs). However, hypertrophy which leads to cartilage loss along the joint space and bone deposition is still remained as a critical limitation. In this study, we fabricated the poly(DL-lactic-co-glycolic acid) (PLGA)/Pluronic F127 scaffolds with PD98059 (bulk loading) and TGF-β2 (surface immobilization) to estimate their potential use as a polymeric matrix for suppression of hypertrophy and enhancement of chondrogenesis from MSCs. The release behaviors of PD98059 and TGF-β2 from the scaffold were quantified using the HPLC and ELISA kit, respectively. The in vitro and in vivo chondrogenesis and hypertrophy behaviors of MSCs in the scaffolds were also investigated.
