Since endotoxin can lead to irreversible shock and death, it is necessary to develop a facile, rapid, sensitive and in situ method to detect it. In this paper, a gold electrode was modified by self-assembled monolayer (SAM) of 3-mercaptopropionic acid (MPA). And a novel DNA aptamer which was as a sensitive function of endotoxin was immobilized on the modified gold electrode by amido bond. The modified electrode was characterized by electrochemical impendence spectroscopy (EIS) in phosphate buffer solution (PBS, pH=7.4). An equivalent circuit model with a constant phase element was used to interpret the obtained impedance spectra. The changes in the charge-transfer resistance were linearly related to the concentration of endotoxin at the range of 0.1~1ng/ml. The activated and modified gold electrodes offers a novel and promising approach to monitor the endotoxin in real-time. |
