Biomimetic 3D scaffold render cells to behave in the similar way that they would do in vivo. In this study, we fabricated hydrogel composed of alginate, gelation and PCL fibrils for better cell cultivation. PCL fibrils were prepared by hydrolysis of PCL nanofibers. To prepare hybrid scaffolds, alginate (Alg) and gelatin (Gel) were respectively dissolved in serum-free DMEM at concentrations of 4% and 10% (w/v), and mixed with 2/1 volume ratio (Alg/ Gel). Following by adding fibrils to Alg-Gel solution with various concentrations, NIH3T3 cells were seeded into fibrils-Alg-Gel solutions and the mixture was crosslinked by calcium chloride for 3h. Elastic modulus of the hybrid scaffolds decreased with the increase of fibrils when no cell was added. However, increasing in fibrils in the scaffolds enhanced the elastic modulus when cells was added. Cell viability study for 14 days also showed that cell viability increased along with the increase of fibrils. |
