Directed evolution of AAV from several rounds of in vitro / in vivo selection is a useful tool to isolate AAV variants with enhanced properties from diverse libraries of capsid mutants. In this study, AAV libraries used for directed evolution are based on wild-type AAVs which have specific tropisms depending on AAV serotype. For directed evolution to be used successfully, it is important to enlarge the diversity of AAV libraries and control mutation rate on AAV capsid during the construction of AAV libraries. Here, several AAV serotype based libraries are generated with Error-prone PCR and have high diversity of the libraries. Also, the mutation rate was controlled by manganese ion concentration. With these AAV libraries, we are conducting in vivo AAV libraries selection on rat nucleus accumbens (NAc) and prefrontal cortex (PFC) to improve transduction efficiency compared to wild type AAV. |
