Optimization of calcitonin(sCT)-sodium tripolyphosphate(STPP) nano ionic complex and its in vitro LLC-PK1 cell permeation, cAMP release in T47D breast cancer cell line and in vivo PK, PD study |
초록 |
sCT-STPP ionic complex was fabricated based on our previous protocols (J controlled Rel 143 (2010) 251–257) and characterized with TEM, DSC and FT-IR. In vitro release test, cAMP release study using T47D breast cancer cell line, LLC-PK1 cell permeation study, in vivo pharmacokinetic and pharmacodynamic studies were performed. The particle size was around 50-100 nm and the complexation efficiency was c.a. 98~99%. sCT was released for 2-3 weeks in vitro. sCT permeation of 1:50 formulation through LLC-PK1 cell was superior than other formulations. The in vivo study has shown that sCT/STPP ionic complexes exhibited superior hypocalcemic effects compared to free sCT. |
