The aim of this study was to develop a new gene carrier consisting of water-soluble chitosan (SC), cystamine(CA), and PEI. We found that cystamine and PEI modified soluble chitosan (SCCAP) can be deliver faster gene into nucleus of mammalian cell. Gelretardation assay showed that pDNA was completely condensed by SCCAP, and protected from DNase. Also, Its transfection was increased by enhanced proton sponge effect with modified PEI, and bioreducible material was dissociated by glutathione of cytosol into cell. In addition, its cellular uptake was investigated by fluorescent microscope. Characterization of SCCAP was analysed by FT-IR, 1H-NMR, DLS and TEM studies. Moreover, Its cytotoxicity was confirmed by MTT assay. These results suggest that SCBP can be apply as a carrier of gene delivery.
