제목 | Fabrication of Cinnamoylated Alginate-Collagen Conduits for Peripheral Nerve Regeneration |
초록 | For surgical repair of a peripheral nerve defect in which direct end-to-end suturing is impossible, various therapeutic strategies have been investigated. Recently, there has been a breakthrough of tubulation methods for nerve repair. In this work, we have designed and fabricated cinnamoylated alginate-collagen nerve guide with a freez drying technique to generate controllable parallel channel in the centeral place. Sponge and tube shaped cinnamoylated alginate-collagen nerve guides were successfully fabricated. Mechanical strength was greatly enhanced after treatment by calcium chloride solution. In this work, eight mice were used for evaluation of peripheral nerve regeneration. The sciatic nerve on the left side was divided in the thigh. The proximal and distal stumps of the severed nerve were inserted into the nerve guide, and the guide was secured to each end by sutures. Eight weeks after implantation, all mice survived and developed no serious surgical complications. In seven mice of experimental group, peripheral nerve was regenerated successfully. Only one mouse has presented bad symptom such as twisting of leg until the end of the observation period. Our newly developed nerve guide is a promising tool for use as a nerve guide tube in peripheral nerve regeneration and provides a suitable experimental model for future clinical application. |
