Molecular Design and D-A Distance Modulation for Conjugated Polyelectrolytes-based FRET DNA detection |

Electrostatic interaction between cationic conjugated polyelectrolytes (CPs) and dye-labeled DNA can be coordinated to give rise to efficient fluorescence resonance energy transfer (FRET) for sequence-specific DNA assays. Same conjugated polyelectrolyte structures with/without molecular spacers are compared in functioning as a donor in FRET-based DNA detection. Fluorescence quenching of the acceptor via electron transfer (eT) in the electrostatic DNA-polymer complex is a main concern in improving the detection sensitivity. This study shows that thermodynamically favorable eT quenching can be controlled kinetically by modulating the D-A intermolecular distance using molecular spacers, which suggests an important molecular design guideline for efficient CPs-based FRET donors. In addition, several approaches to control D-A distance in subnanometer scale will be discussed for the optimized CPs-based FRET DNA detection. |
