We developed a shell cross-linked polymer micelle (SCM) that could preferentially release entrapped drugs in response to intracellular glutathione (GSH). The middle PLys shell of poly(ethylene glycol)-b-poly(L-lysine)-b-poly(L-phenylalanine) (PEG-b-PLys-b-PPha) copolymer micelles was cross-linked with a functional diamine cross-linker containing a GSH-cleavable disulfide bond. In vivo tissue distribution and tumor accumulation of SCMs labeled with the near-infrared (NIR) fluorophore, Cy5.5, were monitored in MDA-MB231 tumor-bearing mice. The cross-linked micelles were accumulated to tumor sites more effectively due to the prolonged circulation in blood, as compared with non-cross-linked micelles. Interestingly, docetaxel (DTX) loaded cross-linked micelles exhibited increased therapeutic efficacy in tumor-bearing mice, compared with free DTX and DTX-loaded non-cross-linked micelles. |
