Self-assembling and Thermo-sensitive Properties of N-acetylated Glycol Chitosan with Controlled N-acetylation |

Glycol chitosan (GC) is a water soluble chitosan derivative proposed as a new material for biomedical applications. With the goal of extending the use of GC in drug delivery system, N-acetylated GCs (AC-GCs) with controlled degree of N-acetylation (DA) ranging from 27.55% to 86.42% were successfully synthesized using acetic anhydride in mild conditions. AC-GCs with DA ranging from 27.55% to 59.90% were soluble in DMSO while native GC is practically insoluble in common organic solvents such as DMSO. Biodegradability of GC was improved by N-acetylation, in which faster biodegradation rate was achieved with higher DA. In aqueous media, AC-GC could self-assemble into spherical nanoparticles with a mean diameter of 187.9 nm and unimodal size distribution. Furthermore, AC-GC with DA of 86.42% dissolved in PBS (7 wt%) exhibited thermo-sensitive sol-gel transition at 37 ºС. The AC-GCs with self-assembling and thermosensitive properties could be useful for various biomedical applications. |
