제목 Enhancement of adhesion and osteogenic differentiation of adipose-derived stromal cells on fibrin/hyaluronic acid coated scaffold with BMP-2
초록 Bone tissue engineering requires well-defined scaffold with highly open pore. Multi head deposition system (MHDS) has raised great interest for fabricating scaffold, since it allows one for highly open and interconnective-porous structure without use of any cytotoxic solvent, permitting diffusion of nutrients and oxygen effectively. However, this method is not suitable for introducing protein due to heating process. In this study, fibrin/hayaluronic acid (HA) coating on the surface of scaffold could overcome the limitation of the scaffold fabricated using MHDS by incorporating bone morphogenetic protein-2 (BMP-2) into hydrogel. The in vitro release of BMP-2 from fibrin/HA-coated scaffold was sustained for 3 days. Furthermore, significantly enhanced initial cell attachment and stimulated alkaline phosphatase activity on fibrin/HA-coated scaffold. The implantation of ASCs seeded on BMP-2 loaded-fibrin/HA-coated scaffolds showed more extensive bone tissue formation and mineralization.
