Stratum corneum plays a critical role in absorption of active ingredient into skin. Previous emulsion has low efficiency of percutaneous absorption and efficacy. In this study, we developed liquid crystal (LC) emulsion which has ordered structure. It has multi lamellar structure which is similar with the structure of intercellular lipids that consisted of stratum corneum. LCE was consisted of free fatty acid, ceramide and cholesterol. Hydrophilic niacinamide and hydrophobic retinyl palmitate was incorporated in the LC for model drug. The structure of LC was characterized by polarization microscope and cryo SEM. Percutaneous absorption and cell proliferation study of LC was performed using artificial membrane, Franz diffusion cell and MTT assay, Pico-green assay. Absorbed drug was quantified by HPLC and the result showed increased absorption of drug and cell proliferation in the case of LCE compared with original emulsion. |
