제목 Tissue Engineering Disc using PLGA/DBP scaffolds
초록 Since there is no optimal treatment for back pain associated with disc degeneration, focus has shifted toward replacement using tissue engineered disc. Demineralized bone particle was obtained bovine and has a powerful inducer of bone and cartilage specialization. We were fabricated natural/synthetic hybrid scaffolds using PLGA and DBP. Scaffold seeded on disc cell was cultured during 4weeks. Morphology of scaffolds and cell proliferation was characterized by SEM and MTT assay. Expressed gene such as aggrecan and type II collagen was confirmed RT-PCR. Influence of DBP content for tissue formation was investigated by H&E, Safranin-O, Masson’s trichrome staining. In SEM results, we observed that all scaffold has uniform porosity. But PLGA/DBP scaffold has good cell viability and attachment. In histology result, Scaffold containing DBP was the more the density of disc cell than PLGA only. Acknowledgements: This research was supported by KMOHW(0405-BO01-0204-0006) and SCRC(SC3100).
