Blood clotting is an important step in hemostasis to stop bleeding. When the skin is damaged, the activated platelets release granules, one of which contains polyphosphate. Polyphosphate plays important roles in blood clotting cascade such as triggering the clot formation, promoting the activation of factor V, and increasing the thickness of fibrin fibers. In addition, the longer length of polyphosphate promotes the faster formation of blood clot. Herein we shows that phosphorylation of chitosan (Chi-P), which is a natural polymer with hemostatic potential and biocompatibility, promotes the hemostasis. To confirm the conjugation and quantify the amount of the conjugated phosphate, Chi_P is analyzed by FT-IR, XRD, 31P NMR, and 13C NMR. The Chi_P samples with various amount of phosphate are applied on the hemorrhage liver of mouse to confirm the effect of phosphorylation to the hemostasis. As a result, time to hemostasis is reduced as the amount of the conjugated phosphate is increased. |
