Advanced chemical modifications of RNAs have long been established as the key technologies in RNA stabilization and preparation system. However, the applications of RNA interference (RNAi) therapeutics still remain limited to the liver. Beyond the liver, we designed several RNA-conjugates to provide safe and effective RNAi therapeutics for cancer treatment. For instance, as endogenous albumin hitchhiking strategy, we suggest carrier-free maleimide-functionalized anti-miRNAs that enable “piggybacking” onto albumin. The functionalized anti-miRNA drugs covalently bind to cysteine-34 of albumin, thus can extended blood circulation lifetime. This strategy allows anti-miRNA drugs to “hitchhike” to the tumor site. Including albumin-piggybacking approach, carrier-free RNA-conjugate delivery system provides a simple, broadly applicable platform strategy for the systemic delivery of RNAi therapeutics with targets outside the liver. |
