We generated a novel AAV based, L-fucose bound viral vector using a cross-linker. There are several reports that fucosylation of carbohydrate antigen 19-9 (CA19-9), a common pancreatic tumor marker, is increased in pancreatic cancer. In this study, we investigate the hypothesis that Fucose-bound AAV will transduce pancreatic cancer cells more efficiently via CA19-9 receptor mediated endocytosis. Our preliminary data suggests most adeno-associated virus(AAV) serotypes have significantly low gene delivery efficiency to pancreatic cancer cells. Even though the most effective treatment of pancreatic cancer is surgical resection, it has many conditions and is not easy. In addition, since pancreatic cancer is known as a cancer which is relatively ineffective in chemotherapy, that has not actively conducted the chemotherapy for a long time. We expect that the results of this study and those of future study can be used for pancreatic cancer cell-selective diagnosis and treatment. |
