A need for rapid detection of airborne fungus including Aspergillus species which is one of the major causes of human allergy and asthma is continuously increasing. To achieve this, specific binding ligands with high thermal and biochemical stabilities is strongly required. In this study, we discovered specific peptide ligands binding to Aspergiilus niger spores by phage display screening. The phage display-identified peptides were synthesized by solid-phase synthesis and purified (purity > 95%). In flow cytometry assay, we confirmed that peptide ligands exhibitspore-specific binding affinity. In addition, multivalent forms of the peptide ligands were proven to have significantly higher affinity. Finally, we demonstrated that the peptide-modified polydiacetylene (PDA) vesicles undergo color change upon treatment with the target spores from blue to red, which shows a great potential of the developed system as rapid colorimetric fungi sensor. |
